BACKGROUND: Water stress is a major danger to crop yield, hence new approaches to strengthen plant resilience must be developed. To lessen the negative effects of water stress on wheat plants, present study was arranged to investigate the role of synergistic effects of biochar, trans-zeatin riboside (t-ZR), and Azospirillum brasilense on soil improvement and enzymatic activity in water-stressed wheat. RESULTS: In a three-replication experiment comprising of four treatments (T0: Control, T1: Drought stress (DS), T2: DS + t-ZR with biochar, T3: DS + A. brasilense with biochar), we observed notable improvements in soil quality and enzymatic activities in water-stressed wheat plants with the application of t-ZR and A. brasilense with biochar. In drought stress, Treatment having the application of A. brasilense with biochar performs best as compared to the other and significant increased the enzymatic activities such as peroxidase (7.36%), catalase (8.53%), superoxide dismutase (6.01%), polyphenol oxidase (14.14%), and amylase (16.36%) in wheat plants. Different enzymatic activities showed different trends of results. Soil organic C, dissolved organic C, dissolved organic N also enhanced 29.46%, 8.59%, 22.70% respectively with the application of A. brasilense with biochar under drought stress condition. CONCLUSIONS: The synergistic action of A. brasilense and biochar creates an effective microbiological environment that supports essential plant physiological processes during drought stress. This enhancement is attributed to improved soil fertility and increased organic matter content, highlighting the potential of these novel strategies in mitigating water stress effects and enhancing crop resilience.

OBJECTIVE: To evaluate the efficacy of dexamethasone as a final intracanal rinse in relieving postoperative pain of teeth with symptomatic irreversible pulpitis. STUDY DESIGN: Randomised controlled trial. Place and Duration of the Study: Department of Operative Dentistry, PIMS, Islamabad, Pakistan, from June 2019 to December 2020. METHODOLOGY: Sixty patients aged 18- 50 years diagnosed with symptomatic irreversible pulpitis were selected according to the inclusion criterion. After obtaining informed consent, root canal therapy (RCT) was initiated under rubber dam. Pulpectomy was done followed by canal preparation. The lottery method was utilised for the division of patients. Group A (experimental) received dexamethasone (4mg/ml in 5ml syringe) as a final rinse, while Group B (control group) recalled after 1 week and asked whether their pain had relieved or not as a yes/no question. After data collection teeth were obturated and permanent restoration was placed. Data were analysed using Chi-square test. RESULTS: The efficacy of dexamethasone as a final intracanal rinse was greater than saline 86.67% and 20.0%, respectively (p < 0.05) in relieving postoperative pain in teeth with symptomatic irreversible pulpitis. CONCLUSION: Dexamethasone was proved to be more efficacious than saline in alleviating postoperative pain when used as a final intracanal rinse after canal instrumentation. The agriculture and food (agrifood) sectors play key roles in the emergence, spread, and containment of antimicrobial resistance (AMR). Pakistan's first National Action Plan (NAP) on AMR was developed to guide One Health interventions to combat AMR through 2017-2022. To improve subsequent iterations, we assessed the implementation of Pakistan's NAP in the agrifood sectors (NAPag) in October 2022, using the Progressive Management Pathway on AMR tool developed by the Food and Agriculture Organization of the United Nations (FAO). The assessment tool addressed four crucial focus areas of the NAPag: governance, awareness, evidence, and practices. Each focus area contains multiple topics, which involve four sequential stages of activities to progressively achieve systematic management of AMR risk in the agrifood sectors. High-level representatives of the NAPag stakeholders provided information for the assessment through pre-event documentary review and workshop discussions. The assessment results showed that Pakistan's NAPag had an overall moderate coverage (59%) of the anticipated activities. Gaps were particularly notable in strengthening governance, good practices, and interventions in non-livestock sectors. Furthermore, only 12% of the evaluated activities were fully executed and documented, consistently remaining at the planning and piloting stages in the livestock sector across all the examined topics. Insufficient attention to non-livestock sectors, inadequate regulation and enforcement capacity, and resource constraints have hindered scalable and sustainable interventions under the current plan. This assessment provides valuable insights to strengthen the inclusiveness and contribution of the agrifood sectors in the next NAP iteration. In the short-to-medium term, strategic prioritization is necessary to optimize the use of limited resources and target the most critical gaps, such as improving awareness among key stakeholders and fortifying regulations for prudent antimicrobial use. In the long term, integration of AMR into the country's broader health, development, and agricultural transformation agendas will be needed to generate sustainable benefits.

Context: Elderly malnourishment remains a largely unexplored area, despite having a significant effect on the level of mortality and morbidity. This study aims to explore the level of malnourishment and identify those at risk of malnourishment among the elderly population presenting to a teaching hospital in Karachi. Aims: To assess nutritional status and its associated factors among the elderly population. Settings and Design: This study was conducted in outpatient clinics of family medicine at a teaching hospital and its outreach primary care centres from December 2020 to December 2021. The study design was cross-sectional. Materials and Methods: The prevalence of malnourishment and at risk of malnourishment with its associated factors were assessed using the Mini-Nutritional Assessment (MNA) scale as a screening tool among the elderly population (65 years and above) presenting to the family medicine OPDs of a teaching hospital. Statistical Analysis: Data were analysed using the Statistical Package for the Social Sciences (SPSS) version 21. Results: A total of 191 patients were recruited into the study, and the majority of them were males (66.5%). Median age and body mass index (BMI) of participants were 70 (IQR = 74-65) years and 24 (IQR = 27-21) kg/m2, respectively. The median MNA scores were 23.5 (IQR = 20.5-26). A total of 92 (48.2%) participants were found to have a normal nutritional status, 81 (42.4%) were found to be at a risk of malnourishment, and 18 (9.4%) were malnourished. On the multivariable regression model, BMI and ability to manage finances were significantly associated with malnourishment. Conclusion: This study shows an alarmingly high rate of the elderly population at risk of developing malnourishment, and financial independence and increasing BMI were associated with a decreased risk of malnourishment.

Satellite remote sensing is widely being used by the researchers and geospatial scientists due to its free data access for land observation and agricultural activities monitoring. The world is suffering from food shortages due to the dramatic increase in population and climate change. Various crop genotypes can survive in harsh climatic conditions and give more production with less disease infection. Remote sensing can play an essential role in crop genotype identification using computer vision. In many studies, different objects, crops, and land cover classification is done successfully, while crop genotypes classification is still a gray area. Despite the importance of genotype identification for production planning, a significant method has yet to be developed to detect the genotypes varieties of crop yield using multispectral radiometer data. In this study, three genotypes of wheat crop (Aas-'2011', 'Miraj-'08', and 'Punjnad-1) fields are prepared for the investigation of multispectral radio meter band properties. Temporal data (every 15 days from the height of 10 feet covering 5 feet in the circle in one scan) is collected using an efficient multispectral Radio Meter (MSR5 five bands). Two hundred yield samples of each wheat genotype are acquired and manually labeled accordingly for the training of supervised machine learning models. To find the strength of features (five bands), Principle Component Analysis (PCA), Linear Discriminant Analysis (LDA), and Nonlinear Discernment Analysis (NDA) are performed besides the machine learning models of the Extra Tree Classifier (ETC), Random Forest (RF), Support Vector Machine (SVM), Decision Tree (DT), Logistic Regression (LR), k Nearest Neighbor (KNN) and Artificial Neural Network (ANN) with detailed of configuration settings. ANN and random forest algorithm have achieved approximately maximum accuracy of 97% and 96% on the test dataset. It is recommended that digital policymakers from the agriculture department can use ANN and RF to identify the different genotypes at farmer's fields and research centers. These findings can be used for precision identification and management of the crop specific genotypes for optimized resource use efficiency.

BACKGROUND: Chromium (Cr) contamination in soil poses a serious hazard because it hinders plant growth, which eventually reduces crop yield and raises the possibility of a food shortage. Cr's harmful effects interfere with crucial plant functions like photosynthesis and respiration, reducing energy output, causing oxidative stress, and interfering with nutrient intake. In this study, the negative effects of Cr on mung beans are examined, as well as investigate the effectiveness of Azospirillum brasilense and salicylic acid in reducing Cr-induced stress. RESULTS: We investigated how different Cr levels (200, 300, and 400 mg/kg soil) affected the growth of mung bean seedlings with the use of Azospirillum brasilense and salicylic acid. Experiment was conducted with randomized complete block design with 13 treatments having three replications. Significant growth retardation was caused by Cr, as were important factors like shoot and root length, plant height, dry weight, and chlorophyll content significantly reduced. 37.15% plant height, 71.85% root length, 57.09% chlorophyll contents, 82.34% crop growth rate was decreased when Cr toxicity was @ 50 µM but this decrease was remain 27.80%, 44.70%, 38.97% and 63.42%, respectively when applied A. brasilense and Salicylic acid in combine form. Use of Azospirillum brasilense and salicylic acid significantly increased mung bean seedling growth (49%) and contributed to reducing the toxic effect of Cr stress (34% and 14% in plant height, respectively) due to their beneficial properties in promoting plant growth. CONCLUSIONS: Mung bean seedlings are severely damaged by Cr contamination, which limits their growth and physiological characteristics. Using Azospirillum brasilense and salicylic acid together appears to be a viable way to combat stress brought on by Cr and promote general plant growth. Greater nutrient intake, increased antioxidant enzyme activity, and greater root growth are examples of synergistic effects. This strategy has the ability to reduce oxidative stress brought on by chromium, enhancing plant resistance to adverse circumstances. The study offers new perspectives on sustainable practices that hold potential for increasing agricultural output and guaranteeing food security.

Cancer has remained one of the leading causes of death worldwide, with a lack of effective treatment. The intrinsic shortcomings of conventional therapeutics regarding tumor specificity and non-specific toxicity prompt us to look for alternative therapeutics to mitigate these limitations. In this regard, we developed multifunctional bimetallic (FeCo) bi-MIL-88B-FC MOFs modified with folic acid-conjugated chitosan (FC) as drug delivery systems (DDS) for targeted delivery of 5-Fluorouracil (5-FU). The bi-MIL-88B nanocarriers were characterized through various techniques, including powder X-ray diffraction, scanning electron microscopy, energy-dispersive X-ray, thermogravimetric analysis, and Fourier transform infrared spectroscopy. Interestingly, 5-FU@bi-MIL-88B-FC showed slower release of 5-FU due to a gated effect phenomenon endowed by FC surface coating compared to un-modified 5-FU@bi-MIL-88B. The pH-responsive drug release was observed, with 58% of the loaded 5-FU released in cancer cells mimicking pH (5.2) compared to only 24.9% released under physiological pH (5.4). The in vitro cytotoxicity and cellular internalization experiments revealed the superiority of 5-FU@bi-MIL-88B-FC as a highly potent targeted DDS against folate receptor (FR) positive SW480 cancer cells. Moreover, due to the presence of Fe and Co in the structure, bi-MIL-88B exhibited peroxidase-like activity for chemodynamic therapy. Based on the results, 5-FU@bi-MIL-88B-FC could serve as promising candidate for smart DDS by sustained drug release and selective targeting.

Cadmium (Cd) pollution is a growing environmental problem that negatively impacts plant growth and development, particularly in maize. In this research, the impact of farmyard manure (FYM) and salicylic acid (SA) on rhizospheric characteristics and the reduction of Cd stress in maize was examined at Government College (GC) University, Lahore, in 2022. The experiment was arranged with a randomized design, including three replications of 12 treatments (T1 = Control; T2 = Farmyard manure; T3 = Salicylic Acid; T4 = 100 mg/kg of soil Cd; T5 = 200 mg/kg of soil Cd; T6 = Farmyard manure + Salicylic acid; T7 = FYM + 100 mg/kg soil Cd; T8 = FYM + 200 mg/kg soil Cd; T9 = SA + 100 mg/kg soil Cd; T10 = SA + 200 mg/kg soil Cd; T11 = FYM + SA + 100 mg/kg soil Cd; T12 = FYM + SA + 200 mg/kg soil Cd). Results demonstrated that Cd stress negatively affected the maize plant and soil properties, but the application of SA and FYM was effective to mitigate the Cd stress up to a certain level. A reduction of 41.52%, 39.14%, and 39.94% in root length, length of the leaf, and crop growth rate was noticed, due to the Cd stress at 200 mg/kg soil, but this reduction was reduced to 18.83%, 10.35%, and 12.26%, respectively, when FYM and salicylic acid were applied as a combined application under the same stress level of Cd. The root biomass, leaf surface area, and length were all improved by SA and FYM, which enhanced the plant's capacity to absorb nutrients and improve growth under Cd stress. In conclusion, the use of salicylic acid together with farm manure can be an effective approach to mitigate Cd stress in maize crops.

Water management techniques are improving at the farm level, but they are not enough to deal with the limited availability of water and increased crop yields. Soil microbes play a vital role in nitrogen fixation, improving soil fertility and enhancing plant growth hormones under drought conditions. Therefore, this study was conducted to investigate the impact of water management combined with Azospirillum brasilense and Rhizobium pisi on wheat crop productivity and soil properties in dry regions. Three water management techniques were compared, normal irrigation as a control (C), deficit irrigation (DI), and partial root drying irrigation (PRD), together with the interaction of plant-growth-promoting rhizobacteria (PGPR). Experiments were conducted with six treatments in total: T1 = C + No PGPR, T2 = C + PGPR, T3 = DI + No PGPR, T4 = DI + PGPR, T5 = PRD + No PGPR, and T6 = PRD + PGPR. The highest grain yield was achieved in the control irrigation treatment using seeds inoculated with rhizobacteria, followed by control treatment without any inoculation, and the lowest was recorded with deficit irrigation without rhizobacteria inoculated in the seeds. However, PRD irrigation resulted in significantly higher plant growth and grain yield than the DI treatment. PGPR inoculation combined with PRD resulted in a 22% and 20% higher number of grains per spike, a 19% and 21% higher grain yield, and a 25% and 22% higher crop growth rate compared to rhizobacteria inoculation combined with the DI system in 2021-22 and 2022-23, respectively. This increase was due to the higher production of growth hormones and higher leaf area index under water-limited conditions. A greater leaf area index leads to a higher chlorophyll content and higher food production for plant growth.

Generating highly dispersed metal NPs of the desired size on surfaces such as porous silica is challenging due to wettability issues. Here, we report highly active and well-dispersed Pd incorporated mesoporous MCM-41 (Pd@MCM) using a facile impregnation via a molecular approach based on hydrogen bonding interaction of a palladium ß-diketone complex with surface silanol groups of mesoporous silica. Controlled thermal treatment of so obtained materials in air, argon, and hydrogen provided the catalysts characterized by electron microscopy, nitrogen physisorption, X-ray diffraction and spectroscopy. Gratifyingly, our catalyst provided the lowest ever activation energy (14.3 kJ/mol) reported in literature for dehydrogenation of NaBH4 . Moreover, the rate constant (7×10-3  s-1 ) for the reduction of 4-nitrophenol outperformed the activity of commercial Pd/C (4×10-3  s-1 ) and Pd/Al2 O3 (5×10-3  s-1 ) catalysts.

The flow of unprocessed sewage through municipal sewers is a great source of water contamination. This study aims to observe the pollutants removal efficiencies of walnut shells as an efficient low-cost adsorbent material compared to gravel materials as an anaerobic filter medium. Two models of the De-Centralized Wastewater Treatment System (DEWATS) were constructed. The wastewater flowing from toilets and handwashing places was connected to anaerobic filters filled with walnut shells and gravel. The efficiency of both filter media in the removal of biological oxygen demand (BOD), chemical oxygen demand (COD), total suspended solids (TSS), total dissolved solids (TDS), nitrate (NO3), and phosphate (PO43), pH and temperature were observed at the influent of the settler tank and then at the effluent of the collection tank (CT). Temperature and pH were within the acceptable limit of wastewater discharge. The results also indicated that the walnut shells filter media was more efficient at removing organic pollutants (TSS 94%, BOD5 88%, COD 85%, Nitrate 57%, phosphate 46%, and TDS 29%) than the gravel (TSS 81%, BOD5 82%, COD 84%, Nitrate 35%, phosphate 38%, and TDS 26%) at the successive stages. The average removal efficiency of the walnut shell was 88% while in the gravel case, it was 83%. The removal efficiency of walnut shell filters was extensively better over the complete experiment compared to gravel filters for the removal of pollutants, representing the high sorption capability of the walnut shell material. The results of this study show that the walnut shells may be a very useful substitute for other conventional fillers for anaerobic treatment in the anaerobic filter of DEWATS.

BACKGROUND: The SARS-CoV-2 pandemic, caused by the novel coronavirus, has posed a significant global health threat since its emergence in late 2019. The World Health Organization declared the outbreak a pandemic on March 11, 2020, due to its rapid global spread and impact on public health. New variants have raised concerns about their potential impact on the transmission of the virus and the effectiveness of current diagnostic tools, treatments, and vaccines. This study aims to investigate the effect of new variants in Pakistani virus strains on human receptors, specifically ACE2 and NRP1. In-silico analysis provides a powerful tool to analyze the potential impact of new variants on protein structure, function, and interactions. OBJECTIVES: The SARS-CoV-2 virus is evolving quickly. After being exposed in Wuhan, SARS-CoV-2 underwent numerous mutations, leading to several variants' emergence. These variants stabilize the interaction of spike protein with human receptors ACE2 and NRP1. The study aims to check the molecular effect of these variants on human receptors using the in-silico approach. MATERIAL AND METHODS: We use in-silico mutational tools to analyze new variants in SARS-CoV-2 and to check the molecular interaction of spike protein with human receptors (ACE2 and NRP1). Genomic sequences of 41 SARS-CoV-2 strains were sequenced using Ion Torrent (NGS) and submitted to the GISAID database. Spike protein of SARS-CoV-2 sequence trimmed and translated into a protein sequence using ExPasy. We used multiple sequence alignments to check for variants in the spike protein of strains. We utilized mutation tools such as Mupro, SIFT, SNAP2, and Mutpred2.3D structures of SARS-CoV-2 spike proteins (wild and mutated) to analyze further the mutations, ACE2 and NRP1 modelled by the ITASSER protein modelling server. Interactions of spike proteins (wild and mutant) analyzed by MD Docking, Simulation, and MMGBSA RESULTS: Variants I210T, V213G, S371F, S373P, T478K, F486V, Y505H, and D796Y were identified in SARS-CoV-2 Pakistani strains' spike protein. Variant Y505H were found to affect protein function. MD Docking, MMGBSA and MD simulation revealed that these variants increased spike protein's binding affinity with human receptors (ACE2 and NRP1). MD simulation revealed that mutated spike protein stabilized earlier than wild when interacting with ACE2 after 40 ns and interaction with NRP1 stabilized after 30 ns for mutated spike protein compared to wild. CONCLUSION: These variants in Pakistani strains of SARS-CoV-2 are increasing the stability of spike protein with human receptors. These findings provide insight into how the SARS-CoV-2 virus evolves and adapts to human hosts. This information may help develop strategies to control the virus's spread and develop effective treatments and vaccines in the future.

BACKGROUND: Recurrence rates of atrial fibrillation (AF) remain high even after complete wide area circumferential pulmonary vein isolation (PVI). In recent years adjunct posterior wall isolation (PWI) has been performed in patients with more persistent forms of AF but the benefits remain unclear. AIM: The objective of this meta-analysis was to evaluate the efficacy of adjunct posterior wall isolation in reducing recurrence rates of AF using cryoballoon ablation (CBA). METHODS: We searched PubMed, Google Scholar, Clinicaltrials.gov and Cochrane CENTRAL. We included studies comparing PVI to PVI + PWI in patients with either persistent or paroxysmal AF (PAF) undergoing CBA. After data extraction and quality assessment of the studies, we assessed recurrence rates of atrial tachy-arrhythmias (AF, atrial flutter, and atrial tachycardia) as well as total ablation time and procedural adverse events. Risk ratio (RR), mean difference (MD), and 95% confidence interval (CI) were calculated using Review Manager. RESULTS: Concomitant PWI exhibited a substantial reduction in the risk of AF recurrence (RR: 0.51; 95% CI: 0.42-0.63, p < .00001), as well as all atrial arrhythmias (RR: 0.58; 95% CI: 0.49-0.68, p < .00001). On subgroup analysis, in patients with only PAF, adjunct PWI resulted in significant reduction in recurrence risk of AF (RR: 0.56; 95% CI: 0.41-0.76, p = .0002) as well. There was no significant difference in adverse events between both groups (RR: 0.90; 95% CI: 0.44-1.86; p = .78), whereas total ablation time was significantly increased in PVI + PWI group (MD: 21.75; 95% CI: 11.13-32.37, p < .0001). CONCLUSION: Adjunct PWI when compared to PVI alone decreases recurrence rates of atrial fibrillation after CBA of patients with persistent as well as paroxysmal atrial fibrillation.

The present study was carried out to investigate and identify bioactive compounds along with antioxidant capacity, total flavonoids and total phenolic contents from two saprophytic Amanita species, i.e., mushrooms A. manicata (Berk. & Broome) Pegler and A. nana Singer. Antioxidant potential was assessed by DPPH (2,2-diphenyl-1-picrylhydrazyl) free radical scavenging and ABTS (2,2'-azino-bis-3-ethylbenzothiazoline-6-sulfonic acid) assay, total phenolics, and flavonoids. Both mushrooms were found to possess antioxidants and wide range of phenolics and bioactive compounds. There was maximum percent inhibition (83.2 ± 0.120%) on DPPH by A. manicata. However, maximum percent inhibition using ABTS was found to be 79.5 ± 0.251% by A. nana. Similarly, A. nana possesses maximum amount of both flavonoids and phenolics i.e., 0.3473 ± 0.0088 mg/100 g of catechin and 0.097 ± 0.0011 mg/100 g of GAE equivalent, respectively. Both mushrooms exhibited a variety of natural compounds such as P-coumaric acid, M-coumaric acid, benzoic acid, ferulic acid etc. Ferulic acid, chlorogenic acid, and cinnamic acid were also detected in A. manicata. A. manicata exhibited best and greater antiradical potential than A. nana due the presence of excessive natural bioactive compounds. From the reported results, it was revealed that both inedible mushrooms could be a potential source of antioxidants and secondary metabolites and might be used for making novel drugs in future by pharmaceutical industries.

The influence of corporate social responsibility (CSR) on employees' positive psychological capital in stressful situations remains unexplored in the literature for small and medium enterprises (SMEs). This study aims to examine how CSR could assist employees in developing psychological capital during the COVID-19 pandemic. In order to understand the aforesaid relationship, an attempt is made to study the mediation effect of affective commitment. Structural equation modelling (AMOS 21.0) was used for data analysis and hypothesis testing on a sample of 545 employees from 356 Malaysian SMEs. The results of this study showed that SMEs' CSR policies helped to improve the positive psychological capital of their employees during the COVID-19 pandemic. Also, affective commitment complementary mediates the relationship between CSR and psychological capital. During the COVID-19 pandemic, SMEs should come up with a consistent way to implement CSR policies and procedures to improve employees' psychological capital and increase their affective commitment toward the enterprise. There are two main contributions to the literature from this study, in addition to enriching previous empirical research on CSR. As a first contribution to the CSR literature, it examines how CSR impacts employees' psychological capital during a pandemic. COVID-19 is one of the recent pandemics that offers an opportunity to examine its effects on employee psychological state. Secondly, the results of the study add to the growing body of empirical research that supports affective commitment's significant relationship with CSR and enhances employees' psychological capital during a pandemic in a developing market.

Formulating learning systems for the detection of real-world anomalous events using only video-level labels is a challenging task mainly due to the presence of noisy labels as well as the rare occurrence of anomalous events in the training data. We propose a weakly supervised anomaly detection system that has multiple contributions including a random batch selection mechanism to reduce interbatch correlation and a normalcy suppression block (NSB) which learns to minimize anomaly scores over normal regions of a video by utilizing the overall information available in a training batch. In addition, a clustering loss block (CLB) is proposed to mitigate the label noise and to improve the representation learning for the anomalous and normal regions. This block encourages the backbone network to produce two distinct feature clusters representing normal and anomalous events. An extensive analysis of the proposed approach is provided using three popular anomaly detection datasets including UCF-Crime, ShanghaiTech, and UCSD Ped2. The experiments demonstrate the superior anomaly detection capability of our approach.

The present study was an attempt to evaluate the antimicrobial and anthelmintic potential of two Amanita species, i.e., A. orsonii and A. glarea, along with their qualitative mycochemical screening. Maceration technique was adopted to make crude extracts in non-polar (petroleum ether and chloroform) and polar (ethanol and distilled water) solvents. Qualitative mycochemical screening revealed the presence of a variety of secondary metabolites like terpenoids, flavonoids, tannins, alkaloids, saponins, and cardiac glycosides. Antimicrobial activities were carried out by using agar well diffusing method against four bacterial and one fungal strain. The antibacterial potential ranged between 4.86 ± 0.088 mm and 34.83 ± 0.166 mm with maximum inhibition zone exhibited by petroleum ether extract against Pseudomonas fluorescens and least potential by distilled water macerate of A. orsonii against the Escherichia coli. The antifungal activity ranged between 14.5 ± 0.288 and 24.76 ± 0.145 mm, with the highest potential provided by chloroform extract of A. orsonii and least capability put forward by A. glarea by petroleum ether extract against Fusarium solanii. Antibiotics and antimycotic discs were used as standard and some of the crude extracts showed bigger zone of inhibition as compared to standard. Different concentrations of ethanolic extracts of both mushrooms were subjected to anthelmintic potential against parasite Haemonchus contortus. All the crude extracts were more potent than standard oxfandazole used. Anthelmintic potential ranged between 9.9 ± 0.057 and 54.93 ± 0.033 minutes for death of parasite, while the paralyzed time ranged between 4.86 ± 0.088 and 24.86 ± 0.088 minutes. From the results obtained it was concluded that both mushrooms can be used as potential source of curative antibacterial, antifungal, and anthelmintic agents against several diseases that might be used in pharmaceutical industries for making medicines and to screen out secondary metabolites in future.

The rapid increase in population growth under changing climatic conditions causes drought stress, threatening world food security. The identification of physiological and biochemical traits acting as yield-limiting factors in diverse germplasm is pre-requisite for genetic improvement under water-deficit conditions. The major aim of the present study was the identification of drought-tolerant wheat cultivars with a novel source of drought tolerance from local wheat germplasm. The study was conducted to screen 40 local wheat cultivars against drought stress at different growth stages. Barani-83, Blue Silver, Pak-81, and Pasban-90 containing shoot and root fresh weight >60% of control and shoot and root dry weight >80% and 70% of control, respectively, P (% of control >80 in shoot and >88 in root), K+ (>85% of control), and quantum yield of PSII > 90% of control under polyethylene glycol (PEG)-induced drought stress at seedling stage can be considered as tolerant, while more reduction in these parameters make FSD-08, Lasani-08, Punjab-96, and Sahar-06 as drought-sensitive cultivars. FSD-08 and Lasani-08 could not maintain growth and yield due to protoplasmic dehydration, decreased turgidity, cell enlargement, and cell division due to drought treatment at adult growth stage. Stability of leaf chlorophyll content (<20% decrease) reflects photosynthetic efficiency of tolerant cultivars, while ~30 µmol/g fwt concentration of proline, 100%-200% increase in free amino acids, and ~50% increase in accumulation of soluble sugars were associated with maintaining leaf water status by osmotic adjustment. Raw OJIP chlorophyll fluorescence curves revealed a decrease in fluorescence at O, J, I, and P steps in sensitive genotypes FSD-08 and Lasani-08, showing greater damage to photosynthetic machinery and greater decrease in JIP test parameters, performance index (PIABS), maximum quantum yield (Fv/Fm) associated with increase in Vj, absorption (ABS/RC), and dissipation per reaction center (DIo/RC) while a decrease in electron transport per reaction center (ETo/RC). During the present study, differential modifications in morpho-physiological, biochemical, and photosynthetic attributes that alleviate the damaging effects of drought stress in locally grown wheat cultivars were analyzed. Selected tolerant cultivars could be explored in various breeding programs to produce new wheat genotypes with adaptive traits to withstand water stress.

BACKGROUND: Antimicrobial resistance (AMR) and irrational antimicrobial usage (AMU) are a global problem, particularly evident in low- and middle-income countries including Pakistan. Our study aimed to assess the knowledge, attitudes and prescribing practices (KAP) of field veterinarians regarding AMR and AMU. METHODS: A questionnaire-based online survey was conducted using Google forms to collect data on AMR and AMU from veterinary practitioners in Pakistan between July 25, 2020, and February 2, 2021. The questionnaire was comprised of four sections: 1) Demographic information of the respondents, 2) Knowledge about AMR, and AMU, 3) Antibiotic prescribing behavior, attitudes and influencing drivers and 4) Practices of respondents about AMU and drug disposal. RESULTS: A total of 545 participants (Male 84%, Female 16%) completed the survey. The respondents' qualification (P = 0.008), gender (P = 0.007) and type of practitioner (P < 0.001) had a significant association with the mean knowledge score about AMR. Most of the veterinarians had never attended any training, symposium, or awareness session on AMR (63%) and methods to detect antibiotic residues (71%). Participating veterinarians frequently included "critically important antimicrobials" in their treatment regimens. Most veterinarians (60%) prescribed antibiotics for viral diseases. Our study showed that about 44% of veterinarians disposed of expired drugs in the garbage. Antibiotics were prescribed as mass medication and for prophylaxis by 59% and 33% of the veterinarians, respectively, while about 60% of the veterinarians prescribed antibiotics starting with double doses in their treatment regimens. Importantly, only 4% of the veterinarians recognized the concept of One Health. Male veterinarians had higher odds of prescribing mass medication (OR=3.07, P < 0.001) and use of antibiotics for disease prophylaxis (OR=2.31, P = 0.002) than females, whereas improper disposal of expired antimicrobials (OR=2.12, P = 0.003) was more common in female veterinarians. Government veterinarians had 2 (OR=1.95, P = 0.016) and 3 (OR=3.05, P < 0.001) times higher odds to prescribe antibiotics prophylactically and for mass treatments, respectively compared to full-time private veterinarians. CONCLUSION: Our study identifies key factors influencing veterinarians' decision about antimicrobial prescription and highlights important shortcomings such as lack of training on rational use of antimicrobials, frequent use of antimicrobials for mass medication and prophylaxis, widespread use of critically important antimicrobials, and improper disposal practices. There is a dire need to improve knowledge of veterinary practitioners regarding AMR and to adopt a One Health approach to rationalize AMU at the national level in line with the regional and international guidelines.

Plastic waste has gained remarkable research attention due to its accumulation, associated environmental issues, and impact on living organisms. In order to overcome this challenge, there is an urgent need for its removal from the environment. Under this menace, finding appropriate treatment methods like biodegradation instead of typical treatment methods is of supreme importance. However, there is a limited review on bio-decomposition of plastics, existing microbial species, their degradation efficacy, and mechanism. From this point of view, this study focused on a brief overview of biodegradation such as influencing factors on biodegradation, existing species for macro- and micro-plastics, and present research gap. Degradation percentage, limitations of existing species, and future recommendations are proposed. Microbial species such as bacteria, algae, and fungi have the ability to decompose plastics but they are unable to completely mineralize the plastics. Meanwhile, there is limited knowledge about the involved enzymes in plastics degradation, especially in the case of algae. Bio-decomposition of plastics requires more stringent conditions which are usually feasible for field application. This work will be a reference for new researchers to use this effective strategy for plastic pollution removal.
